The phrase en route, meaning 'while on the way [towards]' is taken from French. (It is now naturalized English.) It is ALWAYS spelled in the French way, with the preposition en, and never with the English preposition on.

AWE also has a note on the spellings, pronunciations and meanings of the different words at Root - rout - route.
