Hypothesis is pronounced with the stress on a 'short' vowel sound, like that in 'got', in the second syllable, and with the '-th-' like the sound of 'THink' - 'high-PO-the-sis', IPA: /haɪ 'pɒ θɛ sɪs/.

You may want to look at Hypothesis for a note on the plural form; and hypothesises (pronunciation) for a note on the pronunciation of the 3rd person singular of the present tense of the verb 'to hypothesize'.
